Do I still need to add Prime to ro water before putting it into the tank.

No, its not necessary. I think its mostly for detoxifying tap water. You could keep it on hand in case of an ammonia/nitrite spike though

It can be useful for QT tanks ime but that pretty useless on RODI water, or an established tank
